At what age do symptoms of NEUROFIBROMATOSIS start to appear?
I have a friend who thinks that he has this disease and he would like to know when symptoms start to appear. Please answer for both Neurofibromatosis type 1 and type 2.
Answer: To diagnosis Neurofibromatosis - patients must have 2 or more of these for NF type 1:
6 or more cafe au lait macules larger than 5mm at birth or by age three.
2 or more neurofibromas (usually occurs in adolescence)
2 or more Lisch nodules of the iris.
Axillary and inguinal freckling - arm pit and in the crease of the groin
Distinctive bone lesions
For NF II
1. Masses on the 8th cranial nerve (seen by imaging)
2. First degree relative with NF II and either the mass on the nerve, or 2 of certain tumors. I.e NF II - your parent or sibling must also have it.
Most of the time NF I is diagnosed VERY early on; the child develops the multiple cafe au lait spots and the parents bring them in or the doctor notices the freckling during an exam.
NF is usually diagnosed later (in the teen years). Hearing loss is one of the first symptoms. Again, you would have a family member with it.
However NF is a autosomal dominant disease -- if you don't have parents with this disease than it is INCREDIBLY rare to have a child with it.

Answer: Neurofibromatosis is a genetic disorder that can cause tumors to form anywhere on the nervous system at any time, while Proteus Syndrome is a condition which causes abnormal growth of the skin and bones- having nothing to do with the nervous system (as far as I know.) Answer: The two conditions deal with different tissues: NF deals with an overgrwoth of the tissues of the nerves; proteus deals with an overgrowth of soft tissue, such as collagen, bone, etc. The two disorders can certainly lead to a grossly malformed person, but the two are totally different. Even though Elephant Man has been stated to have had NF, in all likelihood, he actually had proteus syndrome.

Neurofibromatosis (NF) is a neurological disorder that although fairly common, is hardly heard of. What it does is cause tumors to grow along the nerves, randomly, in the body or on the skin. It can also cause a lot of other things, ranging from scoliosis and learning disabilities to cancer. It is a genetic disorder, progressive, and has no cure.
As a mom of two kids with NF, I have been working towards bringing awareness to my community. I am just curious as to how many people reading this might actually have heard of NF, or possibly either have NF or know someone who does.

Answer: Neurofibromatosis type 1 and 2 are autosomal dominant disorders. They are both caused by creation of dysfunctional proteins that should be (if functional) preventing the disease. As a result of multiple factors (for example, substitution, deletion, or insertion), the proteins that are created do not serve their purpose of "tumor suppression." Most commonly, the proteins are truncated, as a result of a splice site mutation. mRNA tests are not often done to determine the exact mutation, as hundreds of mRNA mutations have been identified.
The most common (ie: cheapest) tests run on patients with the disease do not allow identification of the exact gene mutation, only a change in the size of the dysfunctional protein created, so it is therefore difficult to determine with any accuracy whether the disease is caused by substitution, deletion, or insertion (as it could be caused by all three).

As well, people with no relatives have this disease.... apparently a mutation.
But this disease isn't all that predictable.
Some of those who indeed have this disease, don't have disfiguring problems. They barely know that they have it, even as they age. Another person with the disease has so many tumors as to have a condition almost not fixable.

Answer: its a genetically inherited disease so it does not have any incubation period. There is no treatment, and you can't avoid it. You can transmit it either - you inherit it.

Could you get NEUROFIBROMATOSIS without it being in your past family genetics?
I was wondering how does that happen? Does it happen from food? Or stress? How can you prevent it from happening?
Answer: If you have a mutation in the gene responsible for NF type 1, which is on Chromosome 17, you can develop it. The gene, which produces a protein called neurofibromin is a pretty big gene, so it can mutate fairly easily. Neurofibromin regulates production of a protein called ras (which is responsible for the growth of tissue) in the nerve sheath. If your gene is mutated, you'll produce lower-quality neurofibromin, which will do a bad job of regulating ras in the nerve sheath (allowing it to grow too much). Unless the NF-1 gene mutated before you were born, you can't develop it. The same basic principle applies to NF-2, which is caused by a gene on chromosome 22. I will note that if you have a first-generation mutation, your symptoms could be extremely mild and you might not notice them into adulthood.

Is there anyone else out there with NEUROFIBROMATOSIS?
If so, have you ever tried holistic medicine or alternative medicine? Do you think accupuncture or accupressure will eliminate the bumps associated with Neurofibromatosis?
Answer: I have NF1, and have a number of neurofibromas at various places of my body. I have had several cut out over the years but as you know, new ones will form from time to time. Since the condition is genetic, I do not think there is an alternative medicine that can help... but I have researched high and low. I share your stress over wishing there was a way to make the bumps go away. Depending on if the bump is more on top of the skin or below, some have found success with laser treatment.. but regardless of how the tumor is removed there is usually a scar. For me, every few years will have the doctor remove the largest or most visible ones. I pray that you can still find peace in that you are of great value and worth as a person despite the imperfections. I wish you the very best.
